Improvement of Multiplication Rate through Organ - Callus Mixture Culture in Gladiolus ¢¥ Topaz ¢¥

Abstract

|
This study was performed to establish the optimal culture conditions for producing multiple shoots from the organ-callus mixture derived from callus by using the liquid shaking culture in gladiolus ¢¥Topaz¢¥. Obtaining the organ-callus mixture from callus was very effective when it was grown in liquid shaking medium containing 0.05 §·¡¤L^(-1) BA. Shoot regeneration took place from organ-callus mixture when it was traps-cultured in solid medium containing 0.1 §·¡¤L^(-1) BA at 30¡É under 16 hours daylength. Based on microscopic examination, meristemoid was clearly observed, which was then developed into shoot primordia in organ-callus mixture. On a basis of these results, it was possible to establish the model for a mass production of multiple shoots from organ-callus mixtures.
